This presentation provides an in-depth overview of inflammation in musculoskeletal conditions, including sprains, strains, osteoarthritis, and cartilage degeneration, emphasizing how inflammation acts as both a necessary healing mechanism and a driver of ongoing tissue damage. The presenters explain the biological cascade triggered by musculoskeletal injury—highlighting the roles of pro-inflammatory cytokines such as IL-1, IL-6, TNF-α, RANKL, and NF-κB—and how chronic inflammation can accelerate muscle breakdown, cartilage degradation, and bone loss. Special attention is given to osteoarthritis, where impaired cartilage repair and reduced levels of key growth factors contribute to progressive joint deterioration.
The presentation then introduces bone morphogenetic proteins (BMPs), first discovered in 1965, as critical regulators of bone and cartilage regeneration. BMPs are described as the only known osteoinductive proteins capable of signaling mesenchymal stem cells to differentiate into osteoblasts and chondrocytes while simultaneously modulating inflammatory pathways. Research is reviewed showing that BMPs down regulate inflammatory mediators such as IL-1 and IL-6, inhibit destructive enzymes like MMPs, and support tissue homeostasis—mechanisms that are particularly relevant given the documented decline of endogenous BMPs with aging.
